【摘要】:ThistestmethodmeasurestheconcentrationofATPpresentinthesample.ATPisaconstituentofalllivingcellsincludingbacteriaandfungi.Consequently,thepresenceofATPisareliableindicatorofmicrobialcontaminationinfuelsystems.ATPisnotassociatedwithmatterofnon-biologicalorigin.ThistestmethoddiffersfromTestMethodD4012asfollows:ByprovidingfortherapiddeterminationofATPpresentinafuel(petroleum)sample,afuelandwatermixturesample,fuel-associatedbottomwatersampleandextracellularATPfreelyavailableinthefueloraqueoussamplematrix;Byprovidingforamethodtocapture,extractandquantifyATPusingself-containedtestdeviceandluminometer;ByprovidingamethodofquantifyingATPpresentinfuelorwatermatricesingenerallylessthan10min;andByprovidingfortherapidseparationoftheATPfromchemicalinterferencesthathavepreviouslypreventedtheuseofATPdeterminationsincomplexfluidscontaininghydrocarbonsandotherorganicmolecules.Thistestmethoddoesnotrequiretheuseofhazardousmaterialsanddoesnotgeneratebiohazardwaste.Thistestmethodcanbeusedtoestimateviablemicrobialbiomass,toevaluatetheefficacyofantimicrobialpesticides,andtomonitormicrobialcontaminationinfuelstorageanddistributionsystems.1.1Thistestmethodprovidesaprotocolforcapturing,extractingandquantifyingtheadenosinetriphosphate(ATP)contentassociatedwith:1.1.1Microorganismsfoundinconventionalliquidfuelswithkinematicviscosities(at40x00B0;C)ofx2264;8mm2x00B7;sx2013;1asdescribedinTableX6.1,1.1.2Microorganismsfoundinfuel-associatedbottomwater,and1.1.3Extracellular(non-cellular)ATPpresentinthesamplematrix.1.2TheATPismeasuredusingapatentedbioluminescenceenzymeassay,wherebylightisgeneratedinamountsproportionaltotheconcentrationofATPinthesample.ThelightisproducedandmeasuredquantitativelyusingdedicatedATPtestpensandadedicatedluminometerandreportedin(instrumentspecific)RelativeLightUnits.1.3Thistestmethodisequallysuitableforuseinthelaboratoryorfield.1.4AlthoughbioluminescenceisareliableandprovenmethodforqualifyingandquantifyingATP,thismethoddoesnotdifferentiatebetweenATPfromdifferentsources,forexample,fromdifferenttypesofmicroorganismsuchasbacteriaorfungi.1.5Forwaterorcapturesolutionsamples,theconcentrationrangeofATPdetectablebythistestmethodis1x00D7;10x2013;11Mto3x00D7;10x2013;8Mwhichisequivalentto1x00D7;10x2013;14moles/mLto3x00D7;10x2013;11moles/mLforwatersamplesorcapturesolution.Assumingtestingonfuelphaseisperformedona500mLvolumeoffueltheequivalentconcentrationsisfuelwouldbe:6x00D7;10x2013;11Mto2x00D7;10x2013;14M.1.6ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.7Thisstandarddoesnotpurporttoaddressallo......
